There is no difference. If you enter a + to start a formula  and then click a
cell you get =+, if you enter = you get =.

I suppose the + is acting as a unary +, in the same way as the - in =- is
acting as a unary minus. A unary plus makes no difference  to anything. A
unary - negates the value following.
